Far from random, evolution follows a predictable genetic pattern

ScienceDaily (Oct. 25, 2012) ? Evolution, often perceived as a series of random changes, might in fact be driven by a simple and repeated genetic solution to an environmental pressure that a broad range of species happen to share, according to new research.

Princeton University research published in the journal Science suggests that knowledge of a species' genes -- and how certain external conditions affect the proteins encoded by those genes -- could be used to determine a predictable evolutionary pattern driven by outside factors. Scientists could then pinpoint how the diversity of adaptations seen in the natural world developed even in distantly related animals.

"Is evolution predictable? To a surprising extent the answer is yes," said senior researcher Peter Andolfatto, an assistant professor in Princeton's Department of Ecology and Evolutionary Biology and the Lewis-Sigler Institute for Integrative Genomics. He worked with lead author and postdoctoral research associate Ying Zhen, and graduate students Matthew Aardema and Molly Schumer, all from Princeton's ecology and evolutionary biology department, as well as Edgar Medina, a biological sciences graduate student at the University of the Andes in Colombia.

The researchers carried out a survey of DNA sequences from 29 distantly related insect species, the largest sample of organisms yet examined for a single evolutionary trait. Fourteen of these species have evolved a nearly identical characteristic due to one external influence -- they feed on plants that produce cardenolides, a class of steroid-like cardiotoxins that are a natural defense for plants such as milkweed and dogbane.

Though separated by 300 million years of evolution, these diverse insects -- which include beetles, butterflies and aphids -- experienced changes to a key protein called sodium-potassium adenosine triphosphatase, or the sodium-potassium pump, which regulates a cell's crucial sodium-to-potassium ratio. The protein in these insects eventually evolved a resistance to cardenolides, which usually cripple the protein's ability to "pump" potassium into cells and excess sodium out.

Andolfatto and his co-authors first sequenced and assembled all the expressed genes in the studied species. They used these sequences to predict how the sodium-potassium pump would be encoded in each of the species' genes based on cardenolide exposure.

Scientists using similar techniques could trace protein changes in a species' DNA to understand how many diverse organisms evolved as a result of environmental factors, Andolfatto said. "To apply this approach more generally a scientist would have to know something about the genetic underpinnings of a trait and investigate how that trait evolves in large groups of species facing a common evolutionary problem," Andolfatto said.

"For instance, the sodium-potassium pump also is a candidate gene location related to salinity tolerance," he said. "Looking at changes to this protein in the right organisms could reveal how organisms have or may respond to the increasing salinization of oceans and freshwater habitats."

Jianzhi Zhang, a University of Michigan professor of ecology and evolutionary biology, said that the Princeton-based study shows that certain traits have a limited number of molecular mechanisms, and that numerous, distinct species can share the few mechanisms there are. As a result, it is likely that a cross-section of certain organisms can provide insight into the development of other creatures, he said.

"The finding of parallel evolution in not two, but numerous herbivorous insects increases the significance of the study because such frequent parallelism is extremely unlikely to have happened simply by chance," said Zhang, who is familiar with the study but had no role in it.

"It shows that a common molecular mechanism is used by many different insects to defend themselves against the toxins in their food, suggesting that perhaps the number of potential mechanisms for achieving this goal is very limited," he said. "That many different insects independently evolved the same molecular tricks to defend themselves against the same toxin suggests that studying a small number of well-chosen model organisms can teach us a lot about other species. Yes, evolution is predictable to a certain degree."

Andolfatto and his co-authors examined the sodium-potassium pump protein because of its well-known sensitivity to cardenolides. In order to function properly in a wide variety of physiological contexts, cells must be able to control levels of potassium and sodium. Situated on the cell membrane, the protein generates a desired potassium to sodium ratio by "pumping" three sodium atoms out of the cell for every two potassium atoms it brings in.

Cardenolides disrupt the exchange of potassium and sodium, essentially shutting down the protein, Andolfatto said. The human genome contains four copies of the pump protein, and it is a candidate gene for a number of human genetic disorders, including salt-sensitive hypertension and migraines. In addition, humans have long used low doses of cardenolides medicinally for purposes such as controlling heart arrhythmia and congestive heart failure.

The Princeton researchers used the DNA microarray facility in the University's Lewis-Sigler Institute for Integrative Genomics to sequence the expression of the sodium-potassium pump protein in insect species spanning three orders: butterflies and moths (Lepidoptera); beetles and weevils (Coleoptera); and aphids, bed bugs, milkweed bugs and other sucking insects (Hemiptera).

The researchers found that the genes of cardenolide-resistant insects incorporated various mutations that allowed it to resist the toxin. During the evolutionary timeframe examined, the sodium-potassium pump of insects feeding on dogbane and milkweed underwent 33 mutations at sites known to affect sensitivity to cardenolides. These mutations often involved similar or identical amino-acid changes that reduced susceptibility to the toxin. On the other hand, the sodium-potassium pump mutated just once in insects that do not feed on these plants.

Significantly, the researchers found that multiple gene duplications occurred in the ancestors of several of the resistant species. These insects essentially wound up with one conventional sodium-potassium pump protein and one "experimental" version, Andolfatto said. In these insects, the newer, hardier versions of the sodium-potassium pump are mostly expressed in gut tissue where they are likely needed most.

"These gene duplications are an elegant solution to the problem of adapting to environmental changes," Andolfatto said. "In species with these duplicates, the organism is free to experiment with one copy while keeping the other constant, avoiding the risk that the new version of the protein will not perform its primary job as well."

The researchers' findings unify the generally separate ideas of what predominately drives genetic evolution: protein evolution, the evolution of the elements that control protein expression or gene duplication. This study shows that all three mechanisms can be used to solve the same evolutionary problem, Andolfatto said.

Central to the work is the breadth of species the researchers were able to examine using modern gene sequencing equipment, Andolfatto said.

"Historically, studying genetic evolution at this level has been conducted on just a handful of 'model' organisms such as fruit flies," Andolfatto said. "Modern sequencing methods allowed us to approach evolutionary questions in a different way and come up with more comprehensive answers than had we examined one trait in any one organism.

"The power of what we've done is to survey diverse organisms facing a similar problem and find striking evidence for a limited number of possible solutions," he said. "The fact that many of these solutions are used over and over again by completely unrelated species suggests that the evolutionary path is repeatable and predictable."

Is obesity irreversible? Timing is key in weight loss

ScienceDaily (Oct. 24, 2012) ? Joint research between the University of Michigan and the Argentina-based National Council of Science and Technology (CONICET) has shed light on one of the most frustrating mysteries of weight loss -- why the weight inevitably comes back.

A novel animal model showed that the longer mice remained overweight, the more "irreversible" obesity became, according to the new study that appeared online ahead of print Oct.24 in the Journal of Clinical Investigation.

Over time, the static, obese state of the mice reset the "normal," body weight set point to become permanently elevated, despite dieting that initially worked to shed pounds, authors say.

"Our model demonstrates that obesity is in part a self-perpetuating disorder and the results further emphasize the importance of early intervention in childhood to try to prevent the condition whose effects can last a lifetime," says senior author Malcolm J. Low, M.D., Ph.D., professor of molecular and integrative physiology and internal medicine.

"Our new animal model will be useful in pinpointing the reasons why most adults find it exceedingly difficult to maintain meaningful weight loss from dieting and exercise alone."

The lead author of the study was Viviana F. Bumaschny, M.D., assistant investigator of CONICET.

Obesity affects more than 500 million adults and 43 million children younger than age 5, while related illnesses are the leading preventable cause of death.

Individuals who are overweight have a much higher risk of type 2 diabetes, hypertension, and cardiovascular diseases.

One of the major strengths of the research was a new model of obesity-programmed mice that allowed weight loss success to be tracked at different stages and ages by flipping a genetic switch that controls hunger.

Turning on the switch right after weaning prevented the mice from overeating and ever becoming obese. Similarly, mice that remained at a healthy weight into young adulthood by strict dieting alone were able to maintain normal weight without dieting after turning on the switch. However, chronically overfed mice with the earliest onset of obesity never completely returned to normal weight after flipping the switch, despite marked reduction in food intake and increased activity.

The new findings may raise questions about the long-term success rate of severe calorie restriction and strenuous exercise used later in life to lose weight, such as the extreme regimens seen in the popular reality television show "Biggest Loser."

"Somewhere along the way, if obesity is allowed to continue, the body appears to flip a switch that re-programs to a heavier set weight," Low says. "The exact mechanisms that cause this shift are still unknown and require much further study that will help us better understand why the regaining of weight seems almost unavoidable."

The findings will be published alongside a corresponding commentary "Tipping the scales early: probing the long-term effects of obesity."

NEW ORLEANS (AP) ? Pop star Kelly Clarkson will ride as celebrity grand marshal when the Krewe of Endymion parade rolls on the Saturday before Mardi Gras in 2013.

Clarkson was the first winner of the Fox reality show "American Idol" in 2002. Her hits include "A Moment Like This" and "Because of You."

Organizers say she'll perform at Endymion's ball at the Superdome after the parade.

Clarkson may be upstaged at the Feb. 9 parade by a float that organizers say will be the largest and most elaborate in Carnival history.

The super float will be 250 feet long and carry more than 200 riders. Its design focuses on Pontchartrain Beach, the amusement park that entertained generations on the New Orleans lakefront before closing in 1983.

(AP) ? Police say one person was critically wounded when a gunman opened fire inside the chapel of a Georgia megachurch just south of Atlanta.

Fulton County Police Cpl. Kay Lester says the suspect got away in a vehicle, and was still at large around 11 a.m. Wednesday after the shooting at World Changers Church International.

The church is one of the nation's largest. It is led by the Rev. Creflo Dollar, its founder and senior pastor. There was no indication that he was injured.

Lester said she didn't know how many shots were fired.

Church officials did not immediately return calls seeking comment.

Iran's top cargo shipping line says sanctions damage mounting

DUBAI/LONDON (Reuters) - Targeted Western sanctions are hurting Iran's vital shipping industry and if the pressure continues its biggest cargo carrier will face increasingly grave problems, the head of the Iranian line said.

Many of Iran's imports, including food and consumer goods, arrive on container, bulker and other ships, but the number of vessels calling at its ports has dived by more than half this year as the United States and European Union tighten the screws.

"If the government was not assisting, (sanctions) would have stopped a lot more of our activity," Mohammad Hussein Dajmar, managing director of the Islamic Republic of Iran Shipping Lines (IRISL), said in an interview published on Sunday.

"If this situation continues, certainly our operations will face serious problems. Shipping has been among the primary objectives of sanctions. The damage has had a significant bearing. More pressure will result in greater damage," Dajmar was quoted as saying by Iranian business daily Jahan-e Sanat.

Executives of other Iranian shipping companies such as the National Iranian Tanker Company have been less willing than Dajmar to speak about the impact of sanctions.

IRISL, has been on a Western blacklist of sanctioned entities for years accused of transporting weapons, which it denies.

CYBER ATTACK

Dajmar said IRISL suffered a cyber attack in August 2011.

"The attack was heavy and showed that they (the attackers) were being supported by powerful sources," he said. "Most certainly, some information was stolen from us but we don't have any secret information."

He said the company had recovered deleted information.

"There was considerable damage. A lot of pressure was imposed on us," he said.

IRISL has tried to dodge sanctions by changing its flags and setting up front companies, the U.S. Treasury and the European Union have said. It remains easy to track a vessel as it keeps a unique International Maritime Organization (IMO) number.

"We cannot access European waters through changing flags, so we do not take the risk," Dajmar said.

"The IMO numbers are fixed and finding out who owns a ship is not a complicated task ... The United Nations has not barred the activity of our ships, this is only from the U.S. and Europe."

Dajmar said out of its fleet of 165 ships, 20 were currently laid up and not trading.

While broader Western sanctions are directed chiefly against Iran's nuclear program, which is suspected of having a military goal, IRISL has been pinpointed because of suspicions that it transports weapons for Tehran.

Dajmar said IRISL did not work for any Iranian military agencies.

"It's been years that we do not work with them. They don't have such large needs and any needs that they do have they produce domestically," he said.

"It's not like during the war years when the amount of weapons was large and to move them they used ships," he added, referring to the conflict with Iraq in the 1980s.

As part of the trade restrictions the European Union has put an embargo on ship insurance provision, which forced IRISL to switch to Iranian insurance providers.

"Using such (European) insurers was much more profitable and after losing them, we had to set up domestic insurers," he said.

"We are now in a war. They wanted to force us to fall down but they didn't succeed. Today, in global markets we lose markets and the world is putting limitations on us. We will continue even if there is only one country working with us."

Iran denies that its nuclear research has military purposes.

President Mahmoud Ahmadinejad has faced growing criticism over his handling of the economy, especially after Iran's rial currency plunged by more than a third in recent weeks.

Dajmar was quoted earlier this month as saying Iran's central bank had blocked $50 million of the company's assets - reflecting the acute shortage of U.S. currency.

"A few years ago our profit was about $500 million. The company has a strong base but if this situation continues our activities will definitely face problems," he told the daily.

The last area that Rochwerg talked about was the area of loyalty programs and prepaid cards.? Rochwerg says there is a very small number of people who are thinking about loyalty programs and how they are dealt with in the estate.? Some people are including them as assets and making sure they go to a specific beneficiary designated through the will.? Most people have not thought about this in any detail.

When I think about my personal situation with my dad and his estate, I don?t know much about what loyalty programs he participates in.? I know he has an Air Miles card because he used it when we went to Boston Pizza but other than that, what does he have? ?How many of these points get lost when someone dies.

Even in my own situation, we have quite a few points.? It would be great if the family took a nice vacation together using those points.? I should let me wife know that?s what I would like the points to be used for if I die.? How would she know if we never had the discussion?

(AP) ? Lebanese protesters erected flaming roadblocks and gunmen roamed the streets Saturday in a city on edge after the assassination of a top security official in a powerful car bomb the prime minister linked to the civil war in neighboring Syria.

The crisis raised a terrifying specter for Lebanese who fear their country could easily plunge back into cycles of violence and reprisal that have haunted it for decades.

Friday's blast in the heart of Beirut's Christian area killed eight people, including the country's intelligence chief, Brig. Gen. Wissam al-Hassan. It was the deadliest bombing in Beirut in four years, shattering the country's uneasy calm.

The government declared a national day of mourning for the victims Saturday, but protesters burned tires and set up roadblocks in anger.

Sharbal Abdo, who lives in the neighborhood where the bomb went off, brought his 6-year-old son, Chris, and 12-year-old daughter, Jane, to see the destruction Saturday.

"They were very afraid yesterday," he said. "They need to face this situation. It may be their future."

Lebanese Prime Minister Najib Mikati on Saturday linked the bombing to al-Hassan's high-profile investigation this summer that uncovered what authorities called a plot by Syria to provoke chaos in Lebanon with bombings and assassinations.

"I don't want to prejudge the investigation, but in fact we cannot separate yesterday's crime from the revelation of the explosions that could have happened," Mikati said at a news conference following an emergency Cabinet meeting.

Mikati, who opponents believe is too close to Syria and the Shiite militant group Hezbollah, offered to resign after the bombing, but was asked by President Michel Suleiman to stay.

Al-Hassan's probe led to the arrest of former Information Minister Michel Samaha, one of Assad's most loyal allies in Lebanon. Samaha, who is in custody, is accused of plotting a wave of attacks in Lebanon at Syria's behest. Indicted in absentia in the August sweep was Syrian Brig. Gen. Ali Mamlouk, one of Assad's highest aides.

Samaha's arrest was an embarrassing blow to Syria, which has long acted with impunity in Lebanon. Syria has powerful allies here, including the Shiite militant group Hezbollah, which now dominates the government.

For much of the past 30 years, Lebanese have lived under Syrian military and political domination.

Damascus' hold on Lebanon began to slip in 2005, when former Prime Minister Rafik Hariri was assassinated in truck bomb along Beirut's Mediterranean waterfront. Syria denied having any role. But broad public outrage in Lebanon forced Syria to withdraw its troops from the country.

The killings of anti-Syrian figures continued for years, however, and Assad has managed to maintain his influence in Lebanon through Hezbollah and other allies.

Now, as the Syrian civil war rages just across the border, Lebanon increasingly is getting sucked in.

Mikati said Saturday he had offered to resign after Friday's car bomb, but said Suleiman asked him not to plunge the country into more uncertainty.

The bombing raised fears that the crisis could unleash Lebanon's sectarian tensions, a dire scenario for a country that endured a devastating civil war of its own from 1975-1990.

U.N. Secretary-General Ban Ki-Moon spoke Saturday with Suleiman and stressed the importance of "dissociating the country from regional events" and in Lebanon's sovereignty, a U.N. spokesman said.

The Syrian unrest has already enflamed tensions here. Many of Lebanon's Sunni Muslims have backed Syria's mainly Sunni rebels, while Shiite Muslims have tended to back Assad. Al-Hassan was a Sunni whose stances were widely seen to oppose Syria and Hezbollah.

Hundreds of Sunni protesters marched in force through downtown Beirut Saturday, placing the blame squarely on Syria and Hezbollah for al-Hassan's killing.

"Hezbollah is a terrorist group!" they shouted.

Police were trying to identify the bombers and find out how they managed to target al-Hassan, an important security figure who traveled under great protection and who likely took more precautions following Samaha's arrest.

"We don't expect to reveal the crime within few hours," police commander Maj. Gen. Ashraf Rifi told Future TV. "The investigation is like a puzzle. You collect the pieces and put them together in a logical way."

Al-Hassan had many potential enemies.

Besides his investigation of Samaha, al-Hassan helped investigate the 2005 assassination of former Lebanese Prime Minister Rafik Hariri, a powerful Sunni figure. An international tribunal indicted four members of Hezbollah for Hariri's killing, although the group denies involvement.

His department also had a role in breaking up several Israeli spy rings inside Lebanon in recent years, Lebanese officials said.

Al-Hassan, 47, who was married with two children, is expected to be buried Sunday next to Hariri's tomb in downtown Beirut. His family arrived in Lebanon on Saturday on a private plane from Paris, where they live.

Lebanon's top Sunni cleric, Grand Mufti Mohammed Rashid Kabbani, condemned the assassination, calling it a "criminal explosion that targets Lebanon and its people." He also called for self-restraint, saying "the criminal will get his punishment sooner or later."

But many Lebanese were seething with anger.

In the eastern town of Marj, protesters tried to storm an office of the pro-Syrian Itihad group. Lebanese soldiers pushed them away, wounding five protesters, security officials said. Dozens of people who marched in protest in the border town of Moqueibleh came under fire from the Syrian side of the border, forcing them to disperse, the officials said.

The highway linking central Beirut with the city's international airport was closed, as well as the highway that links the capital with Syria, the officials said.

In the predominantly Sunni northern city of Tripoli, gunmen were roaming the streets on motorcycles and opening fire in the air.

The army issued a statement urging Lebanese to overcome the crisis and coordinate among themselves in order to give a chance to the "the criminal killers who tried through the crime to incite strife and split the country."

Tribunal raps GDA over auction of green patch - Real Estate India

GHAZIABAD: The Ghaziabad Development Authority (GDA) was ordered by the National Green Tribunal on Friday that it has to follow the ?due process of law? for felling down any tree located over a 250 sqm plot in Raj Nagar which is being allotted for setting up of commercial shops.

The orders were passed by the tribunal on Friday upon a petition filed by RWA members of Raj Nagar opposing allotment of the ?green patch? for commercial purposes.

Residents have alleged in the petition that the land-use pattern of the plot is not clearly mentioned in the master plan and that the GDA has in the past designated several land uses for it. The plot has remained unused for over three decades.

The principal bench of the tribunal, which ordered that it ?may be indicated in the allotment order that the trees standing over the plot should not be felled without following due process of the law?, has also ordered that the auction conducted by GDA in respect of the ?disputed plot? would be subject to the final outcome of the petition.

The auction, as per a notice issued by the GDA last month, was scheduled for October 19. The tribunal has issued a notice to the GDA seeking its response to the allegations levelled by residents.

Residents have alleged that despite several representations made to the GDA, there has been no clear-cut response on the exact nature of land use of the plot. ?The Authority has kept residents in the dark over the nature of land use of this plot,? said RWA member Dr Nira Bhan, senior consultant with a private hospital in Noida.

?All we were told after several queries is that this green patch is the same plot which has been put up for auction,? she added.

Residents have argued that the plot of land is home to around 25 full grown trees including Ashoka, Neem and Teak, and have opposed to their felling to set up commercial shops on the ground that they already have easy access to big markets nearby.

?The Raj Nagar District Centre, an upscale commercial hub, is at a walking distance from our sector. We do not want our locality to be crowded with petty shops at the cost of its green lungs,? added Dr Bhan.
